Overview

Wild-type scIL-12 (WT scIL-12) is a recombinant, single-chain version of the proinflammatory cytokine interleukin-12 (IL-12). It is engineered by fusing the p35 and p40 subunits of IL-12 into a single polypeptide chain to ensure stoichiometric expression and bioactivity of the IL-12p70 heterodimer. Developed by Lyell Immunopharma and Outpace Bio as a component for "armored" T-cell therapies, WT scIL-12 is designed to modulate the tumor microenvironment by enhancing the activation and cytotoxic function of T cells and natural killer (NK) cells. In preclinical studies, it is often used as a benchmark to compare against next-generation constructs, such as tumor-restricted IL-12 (trIL-12), which aim to provide localized activity while minimizing the systemic toxicities historically associated with wild-type IL-12 administration.
