Overview

Wildtype CD19 scFv CAR T cells are an experimental cellular immunotherapy consisting of T cells engineered to express a chimeric antigen receptor (CAR) targeting the CD19 protein, a biomarker found on the surface of B cells. In research conducted at the Cleveland Clinic, these "wildtype" cells serve as the baseline control for evaluating the impact of genetic modifications, such as the knockdown of the inhibitory receptor LRIG1. The CAR construct typically utilizes a single-chain variable fragment (scFv) for antigen recognition. Upon binding to CD19-positive cells, such as Raji lymphoma cells, these CAR T cells undergo activation and exert cytotoxic effects. Studies have shown that the persistence and function of these wildtype cells can be limited by the VISTA-LRIG1 signaling axis, which acts as a T-cell checkpoint that engages VISTA to limit CAR T cell durability.
