This autologous dendritic cell vaccine is an investigational immunotherapy developed by Antwerp University Hospital (UZA) for the treatment of advanced solid tumors, including pancreatic, esophageal, liver, and ovarian cancers. The vaccine is produced by isolating monocyte-derived dendritic cells (moDCs) from a patient and loading them via electroporation with messenger RNA (mRNA) encoding the Wilms Tumor 1 (WT1) antigen and the Interleukin-15 (IL-15)/Interleukin-15 receptor alpha (IL-15Ra) complex. WT1 is a tumor-associated antigen frequently overexpressed in various malignancies. The co-expression of IL-15 and IL-15Ra allows the dendritic cells to trans-present the IL-15/IL-15Ra complex to T cells and natural killer (NK) cells. This mechanism provides a potent co-stimulatory signal that enhances the activation, survival, and anti-tumor effector functions of WT1-specific cytotoxic T lymphocytes, aiming to generate a robust and durable immune response against the tumor.
