WIN-34B is a new herbal medicine formulated from extracts of Lonicera japonica Thunb (Japanese honeysuckle) and Anemarrhena asphodeloides Bunge. It has demonstrated significant analgesic and anti-inflammatory effects in various animal models of pain and osteoarthritis, with efficacy comparable to celecoxib and indomethacin. Mechanistically, WIN-34B inhibits the production of key inflammatory mediators such as IL-6, PGE2, MMP-13, NO, IL-1β, and TNF-α by suppressing their mRNA expression in fibroblast-like synoviocytes from rheumatoid arthritis patients and macrophages. It also inhibits IκB signaling pathways and nuclear migration of NF-κB. Additionally, WIN-34B promotes cartilage protection by enhancing chondrogenic differentiation in progenitor cells derived from subchondral bone—markedly increasing the expression of CD105, CD73, type II collagen, aggrecan—and restoring proteoglycan content. These properties suggest its potential as a disease-modifying osteoarthritis drug (DMOAD) candidate[1][2][4][6][8].
