WIN-54954 is a small molecule experimental drug that acts as a broad-spectrum antipicornavirus agent. It is effective against a wide range of picornaviruses, including human rhinoviruses and enteroviruses such as echovirus 9 and coxsackievirus A9. The compound functions by binding to a hydrophobic pocket within the virion capsid protein VP1, thereby inhibiting viral replication. In vitro studies demonstrated potent activity across most rhinovirus serotypes and several enteroviruses at low micromolar concentrations. In vivo studies in mice showed that oral administration of WIN‑54954 could protect against paralysis caused by coxsackievirus A9 or echovirus type 9 when administered after infection[2][6][7]. The drug was initially developed by Sanofi for infectious diseases but has not advanced beyond early clinical development.
