Withalongolide B is a naturally occurring withanolide, a type of C28 steroidal lactone, isolated from the plant *Physalis longifolia*. It is characterized as a potent anti-proliferative and cytotoxic agent with potential therapeutic applications in oncology. Research conducted primarily at the University of Kansas has identified withalongolide B as a lead compound for the development of novel chemotherapeutic agents, demonstrating significant activity against various cancer cell lines, including head and neck squamous cell carcinoma and melanoma. Its mechanism of action is associated with the inhibition of the Heat shock protein 90 (Hsp90) chaperone machinery, which leads to the degradation of oncogenic client proteins and the subsequent induction of apoptosis in cancer cells.
