withanone

Withanone is a naturally occurring withanolide, a C28-steroidal lactone primarily derived from the medicinal plant *Withania somnifera* (Ashwagandha). It is recognized as a potent inhibitor of the mitochondrial chaperone protein GRP75 (also known as mortalin or HSPA9). By binding to GRP75, withanone disrupts its interaction with various proteins, such as p53, thereby inducing apoptosis in cancer cells or modulating cellular stress responses. Recent research has also identified its role in preventing CD28 internalization in T cells by inhibiting GRP75-mediated macropinocytosis, suggesting potential therapeutic applications in heart failure and immune dysregulation. While widely available as a component of dietary supplements, it is extensively utilized in preclinical research for its anti-cancer, neuroprotective, and anti-inflammatory properties.
