WJ-Exo(S3I-201) is a nanoformulation consisting of the small molecule STAT3 inhibitor S3I-201 encapsulated within exosomes derived from human Wharton's jelly mesenchymal stem cells. This formulation improves the delivery of S3I-201 to tumor cells, enhancing anti-tumor efficacy by more effectively suppressing STAT3 activity and promoting cancer cell apoptosis in preclinical models. Mechanistically, WJ-Exo(S3I-201) downregulates STAT3 phosphorylation, reduces anti-apoptotic Bcl-2 expression, increases pro-apoptotic Bax and caspase-3 expression, and impedes cancer cell migration. It exhibits significant tumor growth inhibition and survival benefit in triple-negative breast cancer (TNBC) mouse models and shows immunomodulatory effects by modulating cytokine (TNF-α, IFN-γ, IL-1β, IL-4) secretion. The formulation is designed as an experimental anti-cancer therapy with improved tumor targeting and reduced toxicity relative to free S3I-201[1][3].
