Drug intelligence / Profile preview
WMS-1410 is a selective negative allosteric modulator of GluN2B subunit-containing N-methyl-D-aspartate (NMDA) receptors. Developed by the University of Münster, it is a structurally modified tetrahydro-3-benzazepine derivative designed to improve receptor selectivity and metabolic stability compared to the prototypical GluN2B antagonist ifenprodil. WMS-1410 has been investigated in preclinical models for its neuroprotective, anti-inflammatory, and cytoprotective properties. In models of multiple sclerosis, it reduces the infiltration of auto-aggressive immune cells into the spinal cord and ameliorates symptoms of neuro-inflammation. Additionally, WMS-1410 has demonstrated protective effects in pancreatic beta cells by counteracting oxidative stress, calcium accumulation, and apoptotic cell death induced by permanent NMDA receptor activation, suggesting potential therapeutic utility in preserving beta cell mass in type 2 diabetes mellitus. It also exhibits analgesic activity in preclinical models of neuropathic pain.
