Overview

WNK-IN-11 is a potent and highly selective allosteric inhibitor of the serine/threonine protein kinase WNK1 (With-No-Lysine [K] kinase 1), with an IC50 of 4 nM for WNK1 and significant selectivity over WNK2 and WNK4 as well as a large panel of over 400 kinases. By selectively inhibiting WNK1, WNK-IN-11 disrupts downstream signaling pathways including those involving OSR1 and SPAK kinases, impacting mechanisms such as the regulation of the NLRP3 inflammasome, ion transport, cellular volume regulation, and potentially vasodilation and cancer cell signaling. WNK-IN-11 is a small molecule used as a chemical probe in biological research to investigate WNK1-mediated processes in immunity (such as inflammasome regulation and NK cell function), endothelial biology, cancer, and other WNK1-associated pathologies. It binds allosterically and acts as an ATP-noncompetitive inhibitor. Its structure is C21H21Cl2N5OS, CAS number 2123489-30-3[1][3][5][7].
