Overview

WNK463 is an orally bioavailable, potent, and selective pan-WNK (With No Lysine) kinase inhibitor. Developed by Novartis, it targets the atypical serine-threonine kinases WNK1, WNK2, WNK3, and WNK4. WNK kinases are primarily known for their role in regulating ion flux across cellular membranes, particularly in the kidney, and mutations in these kinases are linked to hypertension (Gordon's syndrome). Recent research has also identified WNK1 as a dependency in certain cancers, such as TAL/LMO-related T-cell acute lymphoblastic leukemia (T-ALL). WNK463 acts by binding to the ATP-binding site of WNK kinases, which is unique due to the replacement of a catalytic lysine with a cysteine. In preclinical models, WNK463 has demonstrated the ability to induce cell cycle arrest and inhibit leukemia progression by disrupting mitotic spindle control and chromosome segregation.
