Overview

WP-1122 is a small-molecule prodrug of 2-deoxy-D-glucose (2-DG), developed to improve the pharmacological profile and potency of 2-DG. It acts as a glycolysis inhibitor and has demonstrated greater efficacy than 2-DG alone in preclinical models, particularly where tumor cells exhibit high glycolytic activity. The drug is being investigated for its antineoplastic and antiviral properties, including potential use against glioblastoma multiforme (GBM), pancreatic cancer, COVID-19, HIV infections, and viral hemorrhagic fevers. Mechanistically, it inhibits glycolysis by delivering active 2-DG after oral administration and also modulates coronavirus spike glycoprotein function[1][3][4][5]. Developed by Moleculin Biotech in collaboration with the University of Texas MD Anderson Cancer Center, WP-1122 has received FDA orphan drug designation for GBM[1][3]. Clinical studies have shown that oral administration leads to rapid conversion to active 2-DG with dose-dependent delivery; maximum tolerated doses have been established in healthy volunteers[5][6].
