WP1220 is a synthetic small molecule and analog of WP1066 developed primarily for topical treatment of dermatological conditions. Its main mechanism is inhibition of the phosphorylated form of STAT3 (p-STAT3), an oncogenic transcription factor implicated in tumor proliferation and immune suppression. By inhibiting p-STAT3, WP1220 suppresses the growth of cutaneous T-cell lymphoma (CTCL) cell lines and may modulate immune responses in affected skin lesions. The drug has been evaluated as a monotherapy for CTCL—specifically Mycosis Fungoides—and was previously investigated for plaque psoriasis but discontinued due to insufficient efficacy. Development has involved Moleculin Biotech (and its predecessor Moleculin), WPD Pharmaceuticals, Dermin, and originated from research at the University of Texas M.D. Anderson Cancer Center[1][2][3][4][5][6].
