WP1244 is a **novel DNA-binding small molecule anticancer agent** that features a modular structure combining intercalation and minor-groove binding motifs, enhancing selectivity for specific DNA sequences. It is characterized by a polyamide moiety attached to an amino sugar and possesses a high molecular weight (981.3 Da). Preclinical studies demonstrate **exceedingly high potency** (subnanomolar IC50 values) against a range of brain tumor cell lines, notably ependymoma and glioblastoma multiforme, as well as robust brain penetration after systemic administration. Its mechanism of action resides in its ability to bind to long DNA sequences, thereby disrupting tumor cell proliferation. WP1244 is part of a drug development program focused on therapies for primary and secondary brain cancers, including metastatic disease, showing promising activity in models of central nervous system malignancy and selectivity over normal cells. The molecule is described as being up to 500 times more potent than classic DNA-binding agents like daunorubicin. WP1244 was originally developed at MD Anderson Cancer Center and is now being advanced by CNS Pharmaceuticals, with a more soluble mesylate salt derivative, WP1874, also in development for intravenous formulation[4][5][7][10].
