WP1732 is a fully water-soluble, intravenously-administered small molecule inhibitor of phosphorylated Signal Transducer and Activator of Transcription 3 (p-STAT3), a key oncogenic transcription factor implicated in cancer cell survival, proliferation, and immune evasion. Developed through research at The University of Texas MD Anderson Cancer Center and licensed to Moleculin Biotech, WP1732 shows selectively high accumulation in pancreatic tissue and demonstrates potent induction of apoptosis and inhibition of p-STAT3 in pancreatic ductal adenocarcinoma (PDAC) models, with additional potential activity in other solid tumors and hematological malignancies. Preclinical models have shown that WP1732 induces apoptosis in cancer cells, inhibits p-STAT3 nuclear localization, and has synergistic antitumor effects when combined with immune checkpoint inhibitors. IND-enabling toxicology and formulation development activities have been conducted in preparation for human clinical trials targeting pancreatic cancer as the primary indication[1][2][3][5][7][9].
