WRG-28 is a potent and selective, allosteric small molecule inhibitor of discoidin domain receptor 2 (DDR2). It acts extracellularly to inhibit receptor–ligand interactions via allosteric modulation, primarily blocking the collagen-induced activation of DDR2. By targeting DDR2, WRG-28 inhibits tumor cell invasion and migration, suppresses tumor-supporting stromal interactions, and reduces metastatic colonization in models of metastatic breast cancer. Preclinical data also suggest efficacy in rheumatoid arthritis models[1][2][3][5][7][9].
