WRN inhibitor is a small-molecule therapeutic being developed by Nutshell Therapeutics that targets the Werner syndrome ATP-dependent helicase (WRN). Currently in the Lead to PCC (Pre-Clinical Candidate) stage, this drug is designed for the treatment of solid tumors. WRN inhibitors are a promising class of targeted oncology agents that leverage the principle of synthetic lethality; specifically, they are highly effective against cancers with microsatellite instability (MSI-H). In these cells, the inhibition of the WRN helicase leads to extensive DNA double-strand breaks and chromosomal instability, ultimately resulting in selective cell death while sparing normal, microsatellite-stable cells.
