WRSs2 is a live attenuated oral vaccine candidate developed to prevent infection by Shigella sonnei, a major cause of bacillary dysentery (shigellosis). It is derived from the S. sonnei Moseley strain and engineered with deletions in key virulence genes, including virG (icsA), senA, and senB, which reduce its ability to spread within the colonic epithelium and decrease enterotoxicity. The vaccine is designed to elicit both systemic and mucosal immune responses—particularly functional IgG- and IgA-mediated bactericidal activity—following oral administration. Clinical studies have demonstrated that WRSs2 induces robust humoral and mucosal immunity with an acceptable safety profile at doses up to 10^6 CFU in healthy adults. The primary indication for development is prevention of shigellosis.
