Overview

WT CART19 refers to wild-type chimeric antigen receptor (CAR) T cells engineered to target the CD19 antigen. In the context of research from the University of Pennsylvania, it serves as a standard anti-CD19 CAR-T control to evaluate the efficacy of genetic modifications, such as the CRISPR-Cas9 knockout of CD5. These cells are designed to recognize and eliminate CD19-expressing B-cell malignancies, including B-cell acute lymphoblastic leukemia (B-ALL) and non-Hodgkin lymphomas (NHL). The construct typically utilizes a signaling domain (such as 4-1BB or CD28) and a CD3-zeta activation domain to trigger T-cell effector functions upon binding to CD19 on tumor cells.
