Overview

WT-CCL27 CAR-T is a research-grade chimeric antigen receptor (CAR) T-cell therapy that utilizes the wild-type (unmodified) natural ligand CCL27 to target the C-C chemokine receptor type 10 (CCR10). CCR10 is a chemokine receptor frequently overexpressed on multiple myeloma cells and is associated with poor prognosis and relapse. Developed by researchers at the University of California, San Francisco (UCSF), this construct serves as a proof-of-concept and baseline comparator in studies investigating CCR10 as a therapeutic target. Preclinical evaluations have shown that the wild-type CCL27 binder possesses relatively low affinity for CCR10, resulting in minimal anti-tumor efficacy. This observation has driven the development of second-generation mutant CCL27 CAR-Ts (such as W-CCL27 and F-CCL27) with enhanced binding and cytotoxic performance comparable to BCMA-targeted CAR-T therapies.
