The WT1 126-134 peptide is a synthetic peptide vaccine derived from the Wilms' tumor protein 1 (WT1). It has been investigated primarily for treating acute myeloid leukemia (AML) and myelodysplastic syndromes (MDS) in patients who are HLA-A2 positive[1][4][5]. ## Description WT1 126-134 peptide is a cancer vaccine targeting the Wilms' tumor gene product 1 (WT1), which is an embryonic transcription factor highly expressed in AML and various carcinomas. The peptide corresponds to amino acids 126-134 of the WT1 protein and is designed to elicit a specific immune response against WT1-expressing cancer cells[5][8]. In clinical trials, the vaccine is typically administered subcutaneously, often in combination with immunological adjuvants such as: - Granulocyte-macrophage colony-stimulating factor (GM-CSF) as a dendritic cell stimulant - Keyhole limpet hemocyanin (KLH) as a T-helper protein - Sometimes Montanide ISA 51 VG as an emulsifying agent[5][9] The vaccine works by stimulating WT1-specific T cells to target and eliminate cancer cells expressing the WT1 antigen. Clinical studies have shown that vaccination can increase both the frequency and functionality of WT1-specific T cells, which were often present but with limited function prior to vaccination[4][5].
