Overview

**WT1-anchored Bifidobacterium longum 420** is a genetically engineered oral protein vaccine platform, developed by anchoring the Wilms’ tumor 1 (WT1) protein, a tumor-associated antigen, onto the probiotic bacterium *Bifidobacterium longum* 420. This oral vaccine aims to induce intestinal immunity against cancer, particularly showing antitumor activity by activating WT1-specific immune responses. Oral administration of this vaccine in preclinical mouse models demonstrated significant tumor size reduction and activation of CD8+ T cells, along with enhanced activation of dendritic cells and increased production of WT1-specific antibodies. The vaccine uses the mucosal immune system via Peyer’s patches to prime systemic antitumor immunity, suggesting a novel intestinal bacterium-based cancer immunotherapy approach. This platform leverages the natural probiotic properties of *B. longum* 420 alongside engineered cancer antigen presentation to stimulate an effective antitumor immune response.
