The WT1 CBDC vaccine is an investigational cord blood-derived dendritic cell (CBDC)-based cancer vaccine developed by the U-DANCE Tumor Immunology Section at UMC Utrecht. The vaccine is manufactured by isolating CD34+ stem cells from umbilical cord blood, which are then expanded and differentiated into dendritic cells. These cells are matured with proinflammatory cytokines and electroporated with mRNA encoding the full-length Wilms' Tumor 1 (WT1) protein, a zinc finger transcription factor that is highly overexpressed in approximately 90% of acute myeloid leukemia (AML) cases. The vaccine is designed to be administered to pediatric patients following cord blood hematopoietic cell transplantation (CB-HCT). Its mechanism of action involves presenting WT1 antigens to the newly reconstituting immune system to stimulate the expansion of WT1-specific cytotoxic T lymphocytes (CTLs), which can then target and eliminate residual AML cells to prevent disease relapse.
