WT1-CTL refers to cytotoxic T lymphocytes (CTLs) specifically engineered or induced to recognize peptides derived from the Wilms' Tumor 1 (WT1) antigen, a zinc-finger transcription factor overexpressed in various cancers, particularly hematologic malignancies such as acute myeloid leukemia (AML) and myelodysplastic syndrome (MDS)[7][9]. WT1-CTL therapies involve the ex vivo generation of T cells that target WT1-expressing tumors and are infused into patients to induce an anti-tumor immune response. These therapies can be autologous or allogeneic (derived from donor cells), and represent a form of adoptive cell therapy or cell-based immunotherapy. WT1-specific CTLs have been shown to induce remission or reduce tumor burden in preclinical models and early clinical trials, with a manageable safety profile[2][3][6][8]. They work by recognizing WT1 peptides presented on MHC Class I molecules on cancer cells and destroying these cells via cytotoxic mechanisms. WT1-CTL therapy is under investigation primarily as a post-transplant immunotherapy to prevent relapse in high-risk leukemia, but is also explored in various WT1-positive solid tumors[4][6].
