WT1 DNA vaccine is an investigational cancer immunotherapy consisting of plasmid DNA encoding immunogenic epitopes from Wilms’ tumour antigen 1 (WT1), often fused to a foreign helper domain (DOM) to enhance CD4+ T-cell help and overall immunogenicity. It is designed to induce WT1-specific CD8+ and CD4+ T-cell responses against WT1-overexpressing malignant cells in hematologic malignancies such as chronic myeloid leukemia (CML) and acute myeloid leukemia (AML), where WT1 acts as a leukemia-associated antigen rather than a driver oncogene. In phase II studies, intramuscular DOM–WT1 DNA vaccination in HLA-A2–positive CML and AML patients generated measurable WT1-specific T-cell responses in the majority of evaluable subjects with an acceptable safety profile, supporting further development of WT1-targeted DNA vaccines as maintenance or minimal residual disease–directed therapy.
