The WT1 LAMP mRNA dendritic cell vaccine is an autologous cellular immunotherapy developed by Antwerp University Hospital (UZA) for the treatment of Wilms' Tumor 1 (WT1)-expressing malignancies, including malignant pleural mesothelioma and acute myeloid leukemia (AML). The vaccine is produced by electroporating a patient's own dendritic cells (DCs) with messenger RNA (mRNA) encoding the WT1 antigen. To enhance the potency of the immune response, the WT1 sequence is fused to a Lysosomal-Associated Membrane Protein (LAMP) signal sequence. This modification directs the translated WT1 protein to the lysosomal compartment within the dendritic cell, facilitating efficient processing and presentation of WT1 peptides on MHC class II molecules. This mechanism ensures the activation of both CD4+ helper T cells and CD8+ cytotoxic T cells, aiming to induce a robust and durable anti-tumor immune response against cancer cells overexpressing the WT1 protein.
