WT1 mRNA dendritic cell vaccine is an autologous cellular immunotherapy developed by Antwerp University Hospital (UZA) for the treatment of hematological malignancies, primarily acute myeloid leukemia (AML). The vaccine is produced by differentiating a patient's peripheral blood monocytes into dendritic cells (DCs) ex vivo. These DCs are then matured and transfected via electroporation with messenger RNA (mRNA) encoding the Wilms' Tumor 1 (WT1) protein, a zinc-finger transcription factor that is highly overexpressed in AML and other cancers. Upon intradermal administration, the transfected DCs present WT1-derived peptides on their surface, which primes and activates the patient's own T cells to recognize and eliminate residual WT1-expressing leukemic cells. This approach is primarily investigated as a post-remission strategy to prevent relapse in high-risk patients.
