The WT1 mRNA-electroporated dendritic-cell vaccine is an autologous, personalized cancer immunotherapy developed by Antwerp University Hospital (University of Antwerp) in Belgium. It consists of mature dendritic cells derived from the patient's own CD14+ monocytes (obtained via leukapheresis) that are electroporated with mRNA encoding the Wilms' Tumor protein 1 (WT1), a tumor-associated antigen overexpressed in various hematological and solid malignancies. Upon intradermal administration, these engineered dendritic cells present WT1-derived epitopes to the patient's immune system, priming and expanding WT1-specific CD4+ and CD8+ T-cell responses to target and eliminate WT1-expressing tumor cells. The vaccine has been evaluated in clinical trials for several indications, including glioblastoma multiforme (GBM), pediatric high-grade glioma (pHGG), diffuse intrinsic pontine glioma (DIPG), acute myeloid leukemia (AML), and malignant pleural mesothelioma.
