Overview

This investigational autologous dendritic cell (DC) vaccine therapy is being developed by the University Hospital, Antwerp, in collaboration with the University of Antwerp. The therapy involves the collection of a patient's monocytes via apheresis, which are then differentiated into dendritic cells and electroporated with mRNA encoding the Wilms' tumor 1 (WT1) protein. WT1 is a tumor-associated antigen that is frequently overexpressed in pediatric high-grade glioma (pHGG) and diffuse intrinsic pontine glioma (DIPG). Once administered intradermally, these mature dendritic cells present WT1-derived peptides to the patient's T-cells, aiming to stimulate a robust, tumor-specific immune response. The therapy is currently being evaluated in the Phase I/II ADDICT-pedGLIO trial (NCT04911621) as an adjuvant treatment following or in combination with standard-of-care chemoradiation (temozolomide) to target residual or recurring tumor cells.
