WT1 peptide-loaded dendritic cells are a form of cell-based cancer immunotherapy in which dendritic cells are loaded with peptides derived from the Wilms’ tumor 1 (WT1) protein, a well-characterized tumor-associated antigen overexpressed in many hematological malignancies and solid tumors[1][2][4][5]. These loaded dendritic cells are then administered to stimulate a potent WT1-specific T-cell response, mobilizing both CD8+ cytotoxic T lymphocytes and CD4+ Th1 helper cells to target and eradicate WT1-expressing cancer cells, including cancer stem cells resistant to chemotherapy and radiotherapy[1][4]. Clinical studies, primarily Phase I/II, have demonstrated safety and signs of efficacy for this therapy in several cancers (e.g., acute myeloid leukemia, myelodysplastic syndromes, breast carcinoma, pancreatic ductal adenocarcinoma, glioblastoma), with adverse events typically limited to mild fever and injection site reactions[1][2][4][5]. The therapy can be used alone or in combination with chemotherapy, immune checkpoint inhibitors, or after stem cell transplant to enhance anti-tumor immune responses[1][2][3][4].
