WT1 peptide-pulsed dendritic cell vaccine is a cellular immunotherapy developed by the National Cancer Institute (NCI) for the treatment of various hematologic malignancies. The vaccine consists of dendritic cells (DCs), which are potent antigen-presenting cells, that have been loaded (pulsed) with peptides derived from the Wilms' Tumor 1 (WT1) protein. WT1 is a transcription factor that is highly overexpressed in many leukemias and lymphomas but has limited expression in normal tissues, making it an ideal target for cancer vaccines. Once administered, these dendritic cells present the WT1 antigens to the patient's T cells, stimulating a specific cytotoxic T-lymphocyte (CTL) response against WT1-expressing malignant cells. In clinical trials, this vaccine has been evaluated in the post-transplant setting, often in combination with donor lymphocyte infusions (DLI) to enhance the graft-versus-leukemia effect in patients with relapsed or residual disease.
