WT1 peptide-pulsed dendritic cell vaccine is an autologous cellular immunotherapy designed to treat cancers overexpressing the Wilms' tumor 1 (WT1) protein. The vaccine is produced by harvesting a patient's peripheral blood mononuclear cells, differentiating them into dendritic cells (DCs), and loading them with WT1-derived peptides. These pulsed DCs are then injected back into the patient, where they present WT1 antigens to the immune system, specifically activating cytotoxic T lymphocytes (CTLs) to recognize and destroy WT1-positive tumor cells. In clinical studies like the PANC-DC/S1 trial in Japan, this vaccine is being evaluated as an adjuvant treatment following curative resection for pancreatic cancer, often in combination with chemotherapy such as S-1, to prevent recurrence and improve survival outcomes.
