Overview

WT1-specific cytotoxic T cells are an investigational adoptive cell therapy developed by Memorial Sloan Kettering Cancer Center (MSKCC) for the treatment of relapsed or refractory multiple myeloma and plasma cell leukemia. This biologic therapy consists of donor-derived T cells that are sensitized ex vivo against the Wilms' Tumor 1 (WT1) protein, an antigen frequently overexpressed in various hematologic malignancies. The therapeutic process involves generating WT1-specific T cells from the original stem cell donor by sensitizing CD3+ enriched T-cell fractions with autologous antigen-presenting cells (APCs) loaded with WT1 peptides. These cells are then propagated in vitro and administered to patients following a T-cell depleted allogeneic hematopoietic stem cell transplant (alloTCD-HSCT). The goal is to provide a targeted graft-versus-myeloma effect to achieve long-lasting remission while minimizing the risk of graft-versus-host disease (GVHD).
