WT1-TOPAbody is a preclinical tri-specific T-cell engager antibody targeting the Wilms tumor 1 (WT1) peptide presented by HLA-A*02 on tumor cells, together with CD3 and the T-cell co-stimulatory receptor 4-1BB, designed using CtM Bio’s TOPAbody multispecific engager platform to induce potent, selective T-cell–mediated cytotoxicity against WT1/HLA-A02–positive hematologic and solid tumors while minimizing off-tumor effects.[2][4][12][16] WT1 is an intracellular tumor-associated antigen overexpressed in many leukemias and solid cancers but restricted in normal tissues, and WT1-TOPAbody uses a TCR-mimic antibody module to recognize the WT1/HLA-A02 complex with high affinity and specificity, showing strong in vitro and in vivo antitumor activity and reduced binding to off-target peptides and CD34+ hematopoietic stem cells compared with a benchmark WT1-CD3 bispecific antibody (RG6007).[2][4][12][16] The molecule is being co-developed by Biocytogen and CtM Bio as a novel immunotherapy candidate for WT1/HLA-A02–positive malignancies, including acute myeloid leukemia and various solid tumors, with planned advancement into clinical development.[2][16]
