Drug intelligence / Profile preview
WT1126-TCR is an investigational T-cell receptor (TCR) engineered T-cell therapy (TCR-T) designed to target the Wilms' tumor protein 1 (WT1) antigen. Specifically, it is engineered to express a recombinant TCR that recognizes the HLA-A*02:01-restricted WT1126-134 peptide (RMFPNAPYL), which is highly expressed in acute myeloid leukemia (AML) and other hematological malignancies. Developed by researchers at the University Medical Center Utrecht, Memorial Sloan Kettering Cancer Center, and the University of Antwerp, the therapy utilizes CRISPR-Cas9 gene editing to knock out endogenous TCR alpha and beta chains, preventing TCR mispairing and competition, thereby enhancing the expression and functional avidity of the transgenic WT1126-TCR. The therapy is being investigated for the prevention and treatment of relapse in pediatric and adult patients with AML following hematopoietic stem cell transplantation.
