wtc51m TCR is a high-affinity (supraphysiological) T-cell receptor (TCR) engineered to target the NY-ESO-1 (157-165) peptide epitope presented by HLA-A*02:01. Developed by researchers at the Ludwig Institute for Cancer Research and the University of Lausanne, it is utilized in preclinical research to study the effects of TCR affinity on the efficacy and safety of adoptive cell transfer (ACT) therapies. In comparative studies, wtc51m is often used as a benchmark for high-affinity binding against TCRs with computationally optimized "optimal" affinity ranges, such as the DMβ TCR. Research indicates that while wtc51m has very high binding strength, TCRs in the optimal affinity range may offer superior functional outcomes, including better tumor infiltration and control in immunocompetent models.
