WTS-004 is an orally bioavailable, fourth-generation epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor (TKI) developed by Hangzhou Wutongshu Pharmaceutical. It is specifically designed to address resistance mechanisms in non-small cell lung cancer (NSCLC), particularly the EGFR C797S mutation that frequently emerges following treatment with third-generation TKIs such as osimertinib. WTS-004 exhibits potent inhibitory activity against EGFR triple mutations, including Exon 19del/T790M/C797S and L858R/T790M/C797S, while maintaining high selectivity over wild-type EGFR. This selectivity is intended to improve the safety profile by reducing off-target toxicities commonly associated with EGFR inhibition, such as skin rash and gastrointestinal distress. The drug is currently undergoing clinical evaluation to provide a therapeutic option for patients with advanced NSCLC who have progressed on prior EGFR-targeted therapies.
