WTX-1011 is a **first-in-class, conditionally activated INDUCER™ T-cell engager molecule** developed by Werewolf Therapeutics targeting **STEAP1** (Six Transmembrane Epithelial Antigen of Prostate 1), a protein highly expressed in prostate cancer. The drug employs **PREDATOR masking technology**, specifically anti-CD3 masking, to restrict T-cell engager activity to the tumor microenvironment, significantly minimizing peripheral cytokine release syndrome—a common toxicity of conventional T-cell engagers. In preclinical models, WTX-1011 shows a long circulating half-life (close to 100 hours) and anti-tumor efficacy, while maintaining less than 0.7% unmasked/active molecule exposure in the periphery. This design aims to maximize the therapeutic index for patients with STEAP1-positive prostate cancer, where current immunotherapy options are limited[1][2][3][4][7].
