WTX-124 + pembrolizumab is an investigational **combination immunotherapy** for the treatment of advanced or metastatic solid tumors. WTX-124 is a conditionally activated interleukin-2 (IL-2) prodrug rationally engineered as a masked "INDUKINE" molecule. It is designed to selectively release wild-type IL-2 within the tumor microenvironment, thus minimizing systemic toxicity while retaining localized immune activation. Pembrolizumab is a monoclonal antibody that targets and inhibits the programmed cell death protein 1 (PD-1) checkpoint, enhancing T cell–mediated antitumor responses. The combination aims to synergize IL-2–mediated T and NK cell activation (via WTX-124) with checkpoint inhibition (via pembrolizumab) to enhance anti-tumor immunity while limiting toxicity. Developed and currently in early clinical evaluation, it is specifically being tested in patients with relapsed/refractory solid tumors, including melanoma, renal cell carcinoma, cutaneous squamous cell carcinoma, and NSCLC, who have previously received checkpoint inhibitors[1][2][3][5].
