WTX-212 is an erythrocyte-anti-PD1 antibody conjugate developed by Westlake Therapeutics for the treatment of cancer, including solid tumors and hematologic malignancies. The drug consists of anti-PD1 antibodies conjugated to red blood cells (RBCs), which primarily distribute to the spleen and vascular system upon administration. The anti-PD1 component targets and inhibits programmed cell death protein 1 (PD-1), thereby blocking PD-1-mediated downregulation of T-cell activation and proliferation. This mechanism activates the peripheral immune system, remodels the tumor microenvironment by reducing immunosuppressive myeloid-derived suppressor cells (MDSCs) and regulatory T cells (Tregs), while increasing effector CD8+ T cells. Preclinical studies have shown that WTX-212 can overcome resistance to immunotherapy in both responsive and non-responsive tumor models, with significant suppression of tumor growth. Early clinical trials indicate that it is well-tolerated with no treatment-related adverse events greater than Grade 2[2][4][5][7][8].
