WTX-330 is a systemically delivered, conditionally activated prodrug of human interleukin-12 (IL-12), engineered to selectively release fully active IL-12 within the tumor microenvironment. The molecule consists of wild-type IL-12 linked to a high-affinity blockade element via a tumor protease-sensitive linker and includes a half-life extension domain. This design aims to maximize anti-tumor immune responses while minimizing systemic toxicities associated with recombinant IL-12 therapy by blocking IL-12 receptor interaction in normal tissues and activating only in tumors. WTX-330 stimulates both innate and adaptive antitumor immunity, including dendritic cell maturation, Th1 differentiation, and T effector cell amplification. It is being developed for various cancers including solid tumors and lymphomas.
