WTX-921 is a novel, conditionally activated recombinant fusion protein engineered to deliver interleukin 10 (IL-10) specifically to inflamed tissues. Developed by Werewolf Therapeutics, it is designed as an "INDUKINE" molecule that remains inactive in circulation and becomes activated only within the inflammatory microenvironment—such as the colon in inflammatory bowel disease (IBD). This targeted approach aims to overcome the dose-limiting toxicities of systemic IL-10 therapy by ensuring localized activation and minimizing off-tissue effects. Preclinical studies demonstrate that WTX-921 reduces tissue damage and inflammatory cytokine production in mouse models of colitis, prevents weight loss, lowers Disease Activity Index scores, and decreases immune cell infiltration in affected colon tissue. The drug incorporates an optimized IL-10 blocking domain for effective masking outside target tissues and utilizes proprietary PREDATOR technology for selective delivery. Its primary indication is IBD; other inflammatory diseases are also under consideration[1][3][5][6][7][9].
