WTX212A is an investigational recombinant fusion protein designed as an interleukin-15 (IL-15) superagonist. It typically consists of human IL-15 or an IL-15 variant complexed with the sushi domain of the IL-15 receptor alpha (IL-15Rα) chain, a configuration that significantly enhances its stability and biological activity compared to native IL-15. WTX212A acts by binding to the IL-15 receptor beta/gamma (IL-15Rβγ) complex on the surface of immune cells, thereby stimulating the proliferation and activation of cytotoxic CD8+ T cells and natural killer (NK) cells. This mechanism is intended to bolster the body's anti-tumor immune response without the extensive activation of regulatory T cells (Tregs) often seen with IL-2 therapies. Developed by Worg Pharma, WTX212A is currently being evaluated in Phase 1 clinical trials, including investigator-initiated studies at the Second Affiliated Hospital of Zhejiang University, for the treatment of advanced malignancies such as non-small cell lung cancer (NSCLC) and small cell lung cancer (SCLC), particularly in combination with PD-1/PD-L1 monoclonal antibodies.
