WuXiTecan2 is a super-hydrophilic linker-payload platform developed by WuXi XDC for the construction of antibody-drug conjugates (ADCs). It features exatecan, a potent camptothecin-derived topoisomerase I inhibitor, as its cytotoxic payload. The platform is specifically engineered with a hydrophilic linker moiety to mitigate the inherent hydrophobicity of exatecan, which often leads to ADC aggregation and poor plasma stability, particularly at high drug-to-antibody ratios (DAR) like DAR8. Preclinical evaluations of WuXiTecan2-based ADCs, including a trastuzumab-conjugated DAR8 format and a dual-payload format paired with monomethyl auristatin E (MMAE), have demonstrated sub-nanomolar potency against HER2-expressing breast and gastric cancer cell lines, improved plasma stability, and favorable tolerability in cynomolgus monkeys.
