WVE-005

WVE-005 is a stereopure antisense oligonucleotide therapeutic candidate developed by Wave Life Sciences for central nervous system indications, designed to knock down expression of the microtubule-associated protein tau (encoded by MAPT) in the brain and spinal cord. Using Wave’s PN (phosphoryl guanidine–containing) backbone chemistry to enhance potency, distribution, and durability, a single intrathecal dose of WVE-005 in non‑human primates produced broad CNS distribution and greater than 77% reduction of MAPT mRNA at 12 weeks, with approximately 50% knockdown persisting for six months post‑injection.[1][2] As a tau-lowering therapy, WVE-005 is being explored for tauopathies such as Alzheimer’s disease and related neurodegenerative disorders.[1][11]
