WVE-007 is an investigational, long-acting GalNAc-conjugated small interfering RNA (siRNA) therapy developed by Wave Life Sciences. It is designed to silence the INHBE gene (Inhibin βE), a genetically validated target associated with healthy weight loss and improved cardiometabolic profiles. By targeting INHBE messenger RNA, WVE-007 promotes fat burning (lipolysis) while preserving muscle mass, aiming for sustainable weight loss and metabolic health improvements. The drug leverages proprietary oligonucleotide chemistry and GalNAc delivery technology for targeted action in the liver, with the potential for once or twice-yearly dosing. Preclinical studies have shown that WVE-007 can induce significant weight loss without muscle loss and may prevent weight regain after discontinuation of GLP-1 therapies like semaglutide. The first-in-human Phase 1 clinical trial (INLIGHT) began in early 2025 to evaluate safety, tolerability, pharmacokinetics, biomarkers of target engagement, body composition changes, and metabolic health outcomes in adults with overweight or obesity[1][2][4][5][6].
