Overview

WVE-3972-01 is an antisense oligonucleotide drug developed by Wave Life Sciences. It specifically targets and binds to the faulty messenger RNA transcripts containing the GGGGCC hexanucleotide repeat expansion in the C9orf72 gene, which is the most common known genetic cause of familial amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). By binding the mutant RNA, the drug promotes its degradation, reducing abnormal dipeptide repeat protein expression and RNA foci, while preserving normal C9orf72 protein function. The drug leverages Wave’s stereopure chemistry platform, producing molecules with defined stereochemistry to enhance potency and specificity compared to stereorandom mixtures. Preclinical studies demonstrated potent and sustained reduction of pathological markers in animal models[1][7][5][10].
