Overview

WW68 (also known as WW-68) is a preclinical-stage, next-generation MDM2 inhibitor and degrader being developed for the treatment of triple-negative breast cancer (TNBC). Developed by researchers at the University of Houston and the University of Tennessee Health Science Center, WW68 represents a paradigm shift in targeting MDM2 by directly inducing MDM2 protein degradation. Unlike traditional MDM2 inhibitors that block the p53-MDM2 interaction and rely on wild-type p53 to exert anticancer effects, WW68 acts in a p53-independent manner. In preclinical models, including clinically relevant TNBC patient-derived xenograft (PDX) models, WW68 has demonstrated potent anti-tumor activity, inducing G2/M phase arrest and apoptosis, and suppressing tumor growth without causing host toxicity.
