WX-037 is an oral small molecule inhibitor of phosphatidylinositol 3-kinase (PI3K), developed for the treatment of cancer. It specifically inhibits PI3K, thereby preventing activation of the PI3K/protein kinase B (AKT)-mediated signaling pathway. This inhibition can result in decreased tumor cell growth and survival. Preclinical studies demonstrated that WX-037 strongly inhibits AKT phosphorylation and shows promising anti-tumor activity in various cancer cell lines and xenograft models, with greater sensitivity observed in PIK3CA mutant or PTEN null cells and tumors. The drug was initially developed by UCB and later acquired by WILEX for further development. Clinical development included a Phase I trial in patients with solid tumors; however, this trial was terminated due to business reasons[1][2][5][7].
