WX-037 + WX-554

WX-037 + WX-554 is an investigational combination of two small molecule inhibitors: WX-037, a phosphatidylinositol 3-kinase (PI3K) inhibitor, and WX-554, a dual mitogen-activated protein kinase kinase 1/2 (MEK1/2) inhibitor. This combination was developed for targeting oncogenic pathways—specifically, the PI3K/AKT/mTOR and MAPK/ERK pathways—which are commonly altered in solid tumors. By simultaneously inhibiting both PI3K and MEK pathways, the combination aimed to induce synergistic anti-tumor effects, achieving more pronounced tumor growth inhibition than either drug alone. Preclinical and early-phase clinical studies in solid tumors demonstrated non-toxic synergistic growth inhibition and pathway blockade.
